Le profilage récent de l'ADN par José Vouillamoz de l'Istituto Agrario di San Michele all'Adige suggère que les ancêtres de Sangiovese sont Ciliegiolo et Calabrese Montenuovo. La première est bien connue comme une ancienne variété en Toscane, la seconde est une relique presque éteinte de la Calabre, la pointe de l'Italie.
